Kinetic-QCL™檢測是一種定量的，動態(tài)檢測革蘭氏陰性細菌內(nèi)毒素。樣品與鱟試劑（LAL）混合，放在溫育板讀數(shù)器上，溶液變?yōu)辄S色指示測定完 成。Kinetic-QCL™顯色檢測特點用于微量注射藥物，疫苗，抗體和生物制劑檢測。相比于動態(tài)濁度法和凝膠檢測法，Kinetic-QCL™ 不會干擾凝膠形成。檢測靈敏度范圍在0.005 EU/ml - 50.0 EU/ml，是所有鱟試劑（LAL）檢測方法中zui靈敏的。Kinetic-QCL™允許許多樣品在96高通量孔板中同時進行檢測，節(jié)約時間和工作量。
Kinetic-QCL™（50-650U）包含8瓶LAL鱟試劑，2瓶標準內(nèi)毒素，3瓶LAL試劑水。每個試劑盒可以檢測192次。

鱟試劑內(nèi)毒素檢測試劑盒§檢測原理：革蘭氏陰性細菌中內(nèi)毒素激活LAL中酶原，已激活的酶催化無色底物Ac-Ile-Glu-Ala-Arg-pNA裂解為肽段和硝基苯胺（pNA）。pNA可以在溫育板分光光度計405nm讀數(shù)，根據(jù)樣品反應(yīng)時間和標準物內(nèi)毒素反應(yīng)時間可以確定樣品中內(nèi)毒素濃度。

§結(jié)果處理：用微孔板讀數(shù)器測出每孔405nm吸光值(相應(yīng)空白孔作對照)。WinKQCLTM Software自動生成標準物反應(yīng)時間和標準內(nèi)毒素濃度log/log線性相關(guān)。如果相關(guān)系數(shù)值≥0.980，則可以建立多項式模型制作標準曲線， 計算樣品內(nèi)毒素濃度。使用WinKQCLTM Software軟件可以線性回歸自動計算出結(jié)果，具體計算過程參閱說明書。

注意事項：
¡ 所有試驗材料都要確保無內(nèi)毒素，玻璃器皿和材料可以250℃，30min滅菌處理。
¡ 調(diào)節(jié)樣品PH到6.0-8.0
¡ 塑料管不建議用于內(nèi)毒素稀釋

Kinetic Chromogenic LAL Assays 英文簡述：
The Kinetic-QCL™ assay is a quantitative, kinetic assay for the detection of Gram negative bacterial endotoxin. A sample is mixed with the LAL/substrate reagent, placed in an incubating plate reader, and monitored over time (via WinKQCL™ Software) for the appearance of a yellow color.
It is the chromogenic nature of the Kinetic-QCL™ assay that makes it the most appropriate choice for testing small volume parenterals, vaccines, antibiotics and biologicals. The Kinetic-QCL™ assay is less impacted by inhibitory products which may interfere with the clotting mechanism in Turbidimetric and Gel Clot assays. With a sensitivity range of 0.005 EU/ml - 50.0 EU/ml, the Kinetic-QCL™ assay is the most sensitive of the LAL assays. Multiple samples can be processed at one time in this 96-well format resulting in time savings in a busy laboratory. An incubating plate reader is required to run this assay.
